Novel Mixed NOP/Opioid Receptor Peptide Agonists

چکیده

The nociceptin/orphanin FQ (N/OFQ)/N/OFQ receptor (NOP) system controls different biological functions including pain and cough reflex. Mixed NOP/opioid agonists elicit similar effects to strong opioids but with reduced side effects. In this work, 31 peptides the general sequence [Tyr/Dmt1,Xaa5]N/OFQ(1-13)-NH2 were synthesized pharmacologically characterized for their action at human recombinant receptors. best results in terms of NOP versus mu opioid potency obtained by substituting both Tyr1 Thr5 N-terminal portion N/OFQ(1-13)-NH2 noncanonical amino acid Dmt. [Dmt1,5]N/OFQ(1-13)-NH2 has been identified as most potent dual NOP/mu peptide agonist so far described. Experimental data have complemented silico studies shed light on molecular mechanisms which binds active form receptor. Finally, compound exerted antitussive an vivo model cough.

An overview of glucagon-like peptide-1 receptor agonists for the treatment of metabolic syndrome: A drug repositioning

Metabolic syndrome (MetS) is a clustering of several cardiovascular risk factors that include: obesity, dyslipidemia, hypertension and high blood glucose, and often requires multidrug pharmacological interventions. The management of MetS therefore requires high healthcare cost, and can result in poor drug treatment compliance. Hence drug therapies that have pleiotropic beneficial effects may be...

Small-molecule agonists for the glucagon-like peptide 1 receptor.

The peptide hormone glucagon-like peptide (GLP)-1 has important actions resulting in glucose lowering along with weight loss in patients with type 2 diabetes. As a peptide hormone, GLP-1 has to be administered by injection. Only a few small-molecule agonists to peptide hormone receptors have been described and none in the B family of the G protein coupled receptors to which the GLP-1 receptor b...

Cardiovascular Effects of Glucagon-Like Peptide-1 Receptor Agonists

Glucagon-like peptide-1 (GLP-1) is a member of the proglucagon incretin family, and GLP-1 receptor agonists (RAs) have been introduced as a new class of antidiabetic medications in the past decade. The benefits of GLP-1 RAs are derived from their pleiotropic effects, which include glucose-dependent insulin secretion, suppressed glucagon secretion, and reduced appetite. Moreover, GLP-1 RAs also ...

Design and Synthesis of Novel 5-ht2a/2c Receptor Agonists Design and Synthesis of Novel 5-ht2a/2c Receptor Agonists

ABSTRACT The neurotransmitter, serotonin (5-hydroxytryptamine, 5-HT), has been linked to such human mental processes as mood sleep and dreaming, sexual behavior, aggression, depression, anxiety, and hallucinogenesis. Serotonin is considered to be relevant in the pathogenesis of several mental and psychosomatic disorders. By understanding the structure of the receptors serotonin binds to, drugs ...

Design, Synthesis and Pharmacological Evaluation of Novel 2-[2-(2-Chlorophenoxy) phenyl]-1,3,4-oxadiazole Derivatives as Benzodiazepine Receptor Agonists

New derivatives of 2-[2-(2-Chlorophenoxy)phenyl]-1,3,4-oxadiazole as candidates for agonistic effect on benzodiazepine receptors were synthesized. Conformational analysis and superimposition of energy minima conformers of the novel compounds on estazolam, a known benzodiazepine agonist, revealed that the main proposed benzodiazepine pharmacophores were well matched. In pharmacological eval...
